The integration of distinct internalization routes is crucialÌýto determine the fate of plasma membrane (PM) receptors andÌýthe output of their signalling pathways. Contact sites betweenÌýcellular organelles adds a further layer of regulation byÌýcreating microdomains that favour different signalling andÌýmetabolic pathways. These regulatory mechanisms are relevantÌýto the epidermal growth factor receptor (EGFR). We found that,Ìýwhile clathrin-mediated endocytosis (CME) is mainly involvedÌýin EGFR recycling and sustaining signalling, EGFRÌýinternalization through non-clathrin endocytosis (NCE) leadsÌýprimarily to receptor degradation and signal extinction,Ìýrepresenting a crucial safety mechanism to protect cells fromÌýoverstimulation. Internalization via NCE involves theÌýformation of tripartite contact sites between the PM, theÌýendoplasmic reticulum (ER) and the mitochondria, where EGF-dependent localized Ca2+ signalling occurs and propagate. Ca2+Ìýrelease is in turn needed for the fission of NCE vesicularÌýstructures and their release in the cytosol. Thus, NCE, byÌýtargeting EGFRs to degradation, restricts EGFR signalling; itÌýalso exerts a positive role in promoting localized Ca2+Ìýsignalling, possibly activating a wider cellular response andÌýregulating cellular processes that extend beyond thoseÌýassociated with the canonical EGFR signalling pathway. ThroughÌýa combination of mathematical modelling and wet-labÌýexperiments, we are currently elucidating how the integrationÌýof distinct endocytic pathways and inter-organelle crosstalkÌýregulate the EGFR physiological responses and which are theÌýmolecular circuitries involved.
